The complexity and heterogeneity of EVs origin and composition make integrated molecular, structural, and functional studies challenging. However, a comprehensive approach is needed to disentangle the mechanisms by which EVs exert their pathophysiological function and to correlate them to their biomolecular content, structure, and morphology. I am currently working at the investigation of the biophysical characteristics of Extracellular Vesicles (EVs) of different origin as at their cellular internalization mechanisms. Experimental work proceeds in parallel to biological and biochemical characterization and involves the innovative use of techniques as the scattering and reflectometry of X-rays and neutrons (SAXS, SANS, NR), differential scanning calorimetry (DSC) and quartz-crystal microbalance with dissipation (QCM-D). Model membranes with different degrees of compositional complexity are exploited, in the form of large unilamellar vesicles (LUVs), supported membranes and Langmuir monolayers.
